    About 6 months ago i took a patch of moss from a nearby forest, put it in a glass jar and added a bit of water. Now, the original rotifers, tardigrades, nematodes etc. have all died and have been replaced by other micro organisms. The moss is still growing. I took a few drops of the water on the bottom of the jar and put that into another jar, filled it with regular tapwater and added a few slices of apple. Now, about 3 weeks later a lot of Colpodea and smaller organisms have been growing in the 2nd jar. Really fun to keep myself busy. Thank you for making these videos. Stay safe.

    Like easy food source what all can make in home I can say is the medium for bacterial petri dish. Just make one infuzion of little sugar,worm water,salt,vegeta or little soup cube and put in your water sample. For that dont use Agar or gelatine . Most of this kind of food source are used to big jars of pound water collections but works in small ones too. Potatos bits or bannanasare good too. Great videos and put more info Oliver. Dont stay in safe mode whit our microworld around us. ;)

    I have a jar with water and it has been sitting in the balcony for nearly a week. I don't have wheat seeds. Can sesame seeds be used too?

      I never tried it. Separate the water into different samples and then add sesame to one, rice to the other, milk to the third etc.

      StarGazer45 if you want a really nice,
      Booming population of paramecium follow this: get a sample of pond water (12-16oz) with a bit of algae (pluck it yourself, 4-6 tbsps is plenty). Then put in a couple small twigs, dead leafs and a decent amount of lettuce ripped (1-1.5 leafs). Leave the jar open and let sit for 3-5 days and boom. Instant thriving population (it will smell horrible, that’s how you know it’s the good stuff)

    What are your recommendations on the safety measures regarding this kind of culturing method? I've heard hay infusions aren't completely safe given that the bacteria cultured are unknown. Thanks in advance

      The safety measures are simple, keep your culture away from food areas and do not eat, smoke or rub your face when handling it, wash your hands well after handling the culture. After you finish using your slides you can put them on a container with a solution of bleach for a few minutes before cleaning the slides same for the tools you used to handle the sample, a glass dropper works well, if you use tweezers be aware they can corrode on the bleach so just leave them there for a short amount of time and that's mostly it. You can dispose the culture on the drain once you want to get rid of it, wash the container with bleach.
      Edit. I should add, avoid as much as possible to handle the sample with sharp tools, one of the biggest reasons for accidental infections in labs are punctures with needles or blades used to handle the microorganisms. The bleach solution to disinfect your material should be at least 5% bleach (1 part bleach 19 parts water).

      Generally infusions are indeed risky, if too concentrated. Do not overfeed, in this case the ciliates will eat them and keep bacterial count low.

    Do ciliates live in dirt? I've collected some dirt and mixed jt with water, but once I put it under the microscope I didn't see any ciliates. I did see a lot of rotifers and nematodes

    Another great video. I believe both cultures are suggested in my copy of Culture Methods for Invertebrate Animals by Lutz, Welch, Galtsoff, & Needham. A wealth of info there, but you make it most accessible.

    I did it with boiled milk. But first took sample with paramecium on clean glass and using needle split it into droplets. Then selected ones only with paramecium and moved to jar. After week or so I had hundrets of them.

      I see no reason why not but I never tried it. Just a small amount (tip of a knife). Overfeeding is easy becasue it is concentrated.

      @@annfambeck2132 Too much food will cause the number of bacteria to go up too quickly. This reduces the oxygen content and then the paramecia can also not grow.

      I think it's ethier between 20x or 40x(objectives) 10x(eyepiece)
      The camera is a DSLR camera from Nikon
      The microscope is an Olympus CH40

      Between 40x and 200x. Anything more is already difficult because the field of view is so small and the cells quickly move out of the field of view and are difficult to chase.

      Yes, but the bread becomes very soggy and starts to fall apart, and therefore it is difficult to take out again to get to the microogranisms.

      I film myself in front of a "green screen" and I also make a video with the microscope (separately). The video editing program will then remove the green background and replace it with the video from the microscope. Search for "green screen" for instructions on how to do this.

    Let's say I live in the Desert and we have water dishes that sit outside and grow algae in them would this be a sufficient water source for potential micro organisms if I can't travel far? The water dishes sit under a tree we grow and sometimes collect leaves in the summer which adds more plant matter in a way for micro organisms. Would it be possible if I scrapped up some algae from that old water dish during a warm day would I be able to find any micro organisms? Water can't really pond up around here in the desert :(

      The easiest thing is to try it out! If algae are growing, then other microorganisms are most likely also growing.

      @@danielle7760 I can tell you ways how to grow Lepadella ovalis at home..
      1. Pour fresh water into a cut plastic bottle cup
      2. Put in leaves, peas, leek and let stay for 2 months (pour unchlorinated water to prevent drought every month or every 3 weeks cuz evaporation) and put the cut plastic bottle with fresh water and leaves between windows (historic palett windows are recommended, we have historic palett widnows at home.)
      3. Put in after 2 months some types of moss and feed the microbes with leek, peas (might cause water greenish because algae grows and nutrients grow enough to feed Lepadella.
      4. Repeat the same way in 2nd step every 3 weeks, to prevent evaporation.
      5. After around 5 months, and repeating the same way in 2nd way and 3rd way you should observing sample of water. And Lepadella would be your best microfriend, you can move with movement of microscope.
      6. Still feed the rotifiers with peas, leek and basil and repeat the same way in 2nd way and 3rd way, to prevent evaporation. But the water would be more greenish.
      7. The procedure must not be disturbed
