First time was in episode 1, when Asta fought Lebuty. When his grimoire first appeared, all magic in the vicinity disappeared and the entire area turned dark. Although, idk if you could call that a mana zone, since anti-magic is the opposite of magic... Anti-mana zone? lmao
I'd disagree if you rewatch it Yuno still had Lebutys chains around him and he could still use his chain magic, the part where his chain disappeared was him being shocked about asta's anti-magic therefore that wouldn't make sense since. Yuno was still wrapped in his chains + he could still attack and use his magic so this was the first time mana zone was used.

Meaning, Yuno could just blow some strong winds at Asta which will make him unable to even get close to him. Second, Asta can hold his demon form for just 5 minutes, if he can even control it, and Asta cannot even hope to stand a chance against Yuno without it. The fights we have seen Yuno in he barely broke a sweat, meaning that we have surely not seen him in a very hard situation which caused him to use his absolute full power. Yes, Asta can cut through magic, but Yuno is just more powerful than him at this point. That, or they are equal.

If Asta used his demon form he might be able to defeat Yuno before it becomes unusable for a period of time

I think it would’ve been interesting if Yuno had bean permanently blinded by this event and then had to only rely on mana to “see” he could then later learn sensing ki from Yami which would’ve also been unique development for him
