The narrative does not match the video. It’s not a 1:1 ratio of blood to PBS; from what I observe, they use at least three volumes of PBS to dilute the blood. Correct me if I'm wrong.

Phosphate buffered saline. It is a buffer at pH similar to blood so that the cells are happy and don't suffer osmotic shock.
Can this be used to collect both the plasma and mononuclear cells, or will the reagent disturb the contents of the plasma?
