Well technically when you inhale helium your voice is higher because helium is less dense so the sound goes faster which makes it sound higher. So it does work on a wind instrument (even though the guy faked it). We tried with some flute players, it just sounds off xD

It's probably a quirk of his psychology more than anything. Yeah, he's likely very ticklish, but it's probably more the expectation of the tickling that gets him laughing, like that part where he gets started even when he didn't actually get tickled.
Sort of like how you can laugh at a memory of something funny. Sure it was funny the first time, but you know what's funny now ahead of time, so why are you laughing again? He knows, or thinks he knows he's going to be tickled, and he's super ticklish, so he kind of auto-laughs or something.
On a somewhat related note. Some people laugh out of nerves, even when it's detrimental to their situation. Anxious laughter and all that.
