It depends on your climate but around May when the leaves are filling in and before most invasive tree pests attack in full force is best.
Correct.. one I do that helps uptake is to water the tree the day before so it is up taking faster when you apply the injections.

What about lac balsam for the holes?
Just leave the holes alone. If done properly, the tree will decompartmentalize and heal itself as with any other minor wound.
