- Principle: DPPH(1,1-Diphenyl-2-picrylhydrazyl) is a stable free radial with red color(absorbed at 517nm). If free radials have been scavenged, DPPH will generated it's color to yellow. This assay uses this character to show free radical scavenging activity.

Thanks for the methodology. However, the results in the table at 7:53 are not the same as in the final graph. If we use the data available, the R^2 of the adjustment is only 0.843. The inhibition values are also incorrects in relation to the absorbance values. Why?
Because he didn't describe his standard preparation. Calculations are based on results from standard curve which we don't know because he didn't show it

Are you preparing 1M or 0.1mM DPPH...I think it is 0.1mM DPPH you are preparing...
when you were making the solution series..you took 1ml, 2ml, 3ml and 4ml of the sample extract..but how much methanol did you add to it?
he completed it to 10 ml (the sign )

yes it can be in liquid form so that eliminates the whole process of extraction and then you just have to work from the making the series dilution process.

7:54 Calculate Inhibition% by hand i.e. ((0.301-0.284)/0.301)*100%=5.65% not 5.86%. All values of Inhibition% are incorrect.

the question is what was the concentration of the sample you tested... later on you can check what is the parallel concentration of a known molecule (like vitamin e for example)

Hello ahmed, as for the comparison of both, I'll have to suit it up accordingly to the thesis, when it comes to DPPH assessment for some studies both solvent affects the antioxidants, and these would be one of those consideration ; using one of solvent could affects the DPPH environments for reduction, may some solvent makes the DPPH unstable as it does when reacting with the antioxidants vice versa ; solubility of both solvent should be different, thus affecting the absorbance. Hopefully these answer could useful for you.
