This video demonstrates the 'Bacterial CRISPR and Fluorescent Yeast Combo kit' from the company The Odin. The Yeast is engineered using a plasmid, so you get to see a more conventional method along with Crispr Cas9.

OMG thank you SO much for making this! I ordered the ODIN kit for my kids and I to do and we could not for the life of us figure out how to do it based on the instruction booklet. THIS was what we needed! I know that took a ton of time to create but this family is SO grateful you took the time to do it! Much appreciation!

Just a a couple of things... Perhaps, you have a lot of bacterial colonies in your plate of LB/Strep because Strep degraded after microwaving (like Kanamycin in your YPD plates). So, you can´t tell that the CrisprCas worked.
Next time use a control plate for bacterial cells without CrisprCas plasmids... if you see colonies in these plates there is something wrong with the Streptomycin plates.

Thanks. I used the Adobe Creative Cloud suite, so photoshop, after effects and I edited this one on Davinci Resolve, more as a test, I normally edit on Premiere. I also created some of the graphics drawing on the iPad with the adobe apps.

@@Jimiticus I have genetic polycystic kidney disease (pkd) by mutations. These pkd genes called faulty genes. So my question is for you can it cut faulty gene by Crispr-cas9 protocol. May Crispr-cas9 is there only one gene protocol and use for all faulty genetics diseases thanks

The kit? It might be, you can send them an email to double check. I'm in Canada and was able to get the kits. It took awhile but there wasn't any problems with the shipping.

Why not just re-culture the one colony that did take up the plasmid containing the GFP and antibiotic resistance gene?
You could've just picked out the glowing patch and placing it on a new agar plate to let only the glowing patch to duplicate so it could've been a success with a little more patience. Still good work dude.
Yeah I wish I did that afterwards, but I wanted to have a successful experimental from beginning to end, my goal wasn't just the glowing yeast for its own sake

I guess it depends on what you want changed! I think the first treatment using Crispr has been approved for some sort of auditory genetic disorder. So it might be awhile!
