Hi, how to complete Keakoon in Mine (3) quest? do I really need to kill Elite Keakoon Guard Leader to obtain the Keakoon Chief Sign Number?

yes and sometimes more than 1 time(s) but that is a party quest so you can team up with other players for easier and faster kill

No. you just have to stand there and the quest will magically work.
