    0:55 "CT value was low". I think Vincent probably means his CT value was _high_, indicating that a large number of cycles was required to get to the positive threshold, and hence the amount of virus indicated by the test was low.

      @@victor_venema You know nothing about me Victor, that couldn't be further from the truth. Anyone interested, the last claim was about the tendency of vaccine recipients getting Bells palsy. I asserted there is a correlation between the Pfizer vax. In a 20,000 person study (20k vaccinated 20k placebo) 4 of the vaccinated group developed Bells (one thankfully got over it) 0 of the placebo group got bells.
      I falsy claimed that is what snopes said, I misread snopes and for that Victor assumes my lives goals and motivations :x
      more will die from lockdowns. 3rd world starvation, Harvard study over 100m dead if lockdowns do not end.

      @@hamiltoncannagar636 I believe here in the UK we're over the worst of it and yet the restrictions are becoming more strict. On one hand the government is imposing these restrictions, on the other hand they're telling everyone they're not working. Hospital admissions and deaths from respiratory infections are at the average level for this time of the year but everywhere is going up a tier everytime they review it. The government and a lot of governments around the world are acting in their personal interests now, here Labour aren't any better they wanted even more restrictions. I wonder after all the vulnerable have been vaccinated what the next excuse to take your rights away will be?

    22-1-2021 11:29am Dear Professor , is it possible that due to the limit of detection level of the serological test used was unable to pick up the antibodies ? It is hard to have comparative testing on covid-19 antibodies . Another question : what is the probability of a mucosal spray vaccine for covid-19 ? And an oral vaccine to cover the gut covid-19 levels ? I am not sure about urinary vaccines or ones that target the intestines . Would the Pfizer vaccine (intramuscular injection ) be able to induce and produce mucosal antibodies ? Once the covid-19 manages to lodge itself in the lungs - the reality is that covid-19 may never be eradicated from the body even with vaccinations ? Thank you .

    Doc you only know how the test works conceptually. Think about how the test is made and attempt to “conceptually” reverse engineer it. All the antibodies are completely unspecific.

    I have high hopes with the new administration making these self administered tests widely available will be a high priority. If everyone at risk just tested twice a week for 2 weeks, then nearly all infectious people would be caught. They isolate for a week until not infectious and dead ends the Ro chain reaction. Also all those who were close to the infectious person could test daily for 14 days before meeting anyone and those infectious people would be caught before they spread it to anyone. Infections would drop 90% every 2 weeks and soon most regions would no longer be at risk.

      The problem with testing daily x 14 days is that the current home tests for nasal swabbing costs about $12 per kit (Binax now) AND shipping can take a week before it gets to your home due to supply & demand.

      Joyful Stand i got Abbott test kits at Walmart off the shelf. $20 for 2 tests. Results in 15 minutes. If the government made them freely available, we could end this pandemic quickly.

    the other sad thing is that u needed to state the % of false negatives , because u know anti-science types and anti-maskers will use this to say that ALL the stats and #s are too high ,when in actuality we all know that there are thousands of people who had covid that didn't get tested . I know of at least 5. they didn't get tested because they were afraid of how it would affect their health insurance (another pitiful amerikan failure )

    It is unfortunate our authorities have not made developing this type of test a real priority. Mass at-home testing could go far toward reducing the spread of infection and making our schools and businesses able to return to more normal operations.

      It's Dr. Mina from Harvard that has been championing the idea that as a "public health" instrument (not a "medical" instrument), these rapid paper antigen tests should be approved. If the FDA were a person, I'd say they needed to be charged with manslaughter for holding back this technology from use. Thousands of people have died needlessly because they didn't/don't allow tests that pick-up high viral load (Ct measures), instead, choosing to compare it to PCR tests directly (sensitivity, specificity) without taking into account that the PCR tests take time...time when the person is spreading the disease.

    I was told the problem with the antibody test is that it’s not very sensitive and if you had a CoVid infection more than a month ago it won’t work. It seems like it’s in the interest of the panic crowd to have an overly sensitive PCR test and a low sensitivity anti - body test.

    Damning to the PCR test. How many false positives are out there?

      I personally know 6 people who tested positive.
      All had these things in common
      -100% asymptomatic
      -no sickness during 2020
      -no residents of their households became ill during home-isolation.
      In fact I know of no one who've had any form of influenza during 2020, including myself.
      I'm from Denmark and we actually count total number of deaths per year, and have done so for a long time.
      2020 has been a year of very low mortality, compared to previous years, there is no sign of a dangerous pandemic. Yet we're locking down yet again because of these bogus numbers given by pcr tests.
      In 2021 and ongoing the consequences will start to show, we've stopped screening and diagnosing cancer and a broad range of other diseases and conditions, and people will inevitably start dying from easily treatable diseases, so it should be expected that the death rates starts climbing.
      Also now when a majority of the population suddenly decides to take a syringe which is not a vaccine, this is mRNA gene-therapy (cov-vacination)
      So that should also cause a huge amount of unexpected death and illness. This will all be attributed to covid19 virus, which will cause even more forced vaccination, forced isolation, and a true pandemic unfolds.

      You may have had low mortality because of the lockdown, Denmark certainly seems to have had less than neighboring Sweden which had no lockdown. My own feeling is that the lives saved during the first wave will be lost during this winter. And there could be other reasons for Sweden's high mortality, after all Malmo took about the same hit as Copenhagen, suggests to me that it's geography, not politics that's driving this thing.

      The obvious conclusion is that there isn't a pandemic. But as they say, the only way to stop this 'pandemic' is to turn off your TV, since this is the only place where the pandemic exists.

    Clearly 1 test was wrong, but you cannot possibly know which one.
    You actually stated at the start, you were sent the antibody test, by a company DEVELOPING the kit.

    People may soon be required to submit to weekly PCR tests, to keep their jobs. Researchers claim that AGnp's (silver nanoparticles) inhibit DNA replication. Question: Can a nasal spray solution of ionic silver and AGnp, administered before a nasal swab sample is taken, defeat a deliberately inflated CT-PCR test?

    Where can you buy at home antibody tests annonomously?
    I have been looking into this for months. I can't find a place that does not require your personal information.
    I just want an annonomous antibody test? But there isn't one available that's odd I think... Kind of makes it seem like this whole covid thing is all about collecting people's info and sending results to govt when you can't just buy one annonomously.

    Since symptoms of an infection are caused by your body fighting that infection; wouldn't "asymptomatic infection" mean you are going to die because an infection is progressing without a response to keep it in check? Is there a mechanism for a viral infection to suddenly disappear without your body fighting it off?

    Unless something very recently changed, these lateral flow tests (although seemingly “point-of-care” or DYI) are actually to supposed be done and interpreted by CLIA licensed labs under the FDA EUA’s so that those labs can collect validation data and report back to the FDA. We were very excited about using them early on in the pandemic as a surrogate for detecting exposure/ immunity when PCR testing wasn’t as readily available for everyone, but their real-world reliability proved to be all over the map depending on the manufacturer. Some claiming a whole blood drop like you used was not adequate and had to be a serum sample for accuracy. Also, they don’t specifically test for neutralizing antibodies and have cross-reactivity with non-SARS-coV-2 coronaviruses. Maybe Dr Griffin and others can help clarify some of these points? Always appreciate your intelligent discussions!!

      Pregnancy tests are routinely done at home with no issues. It is highly regulated, but there is no reason it should be.

      @@loveforsberg530 I think because pregnancy tests were not released in a hurry in response to a deadly pandemic. They are simple but went through usual regulatory measures. There were hundreds of companies outside of the US releasing Covid serology tests without any vetting except by the company themselves... then FDA stepped in appropriately. It was like the Wild West.

      If you use the tests as a *public health* tool, it doesn't matter much if the results are "perfect". The spit or snot test that's being championed by Dr. Mina will keep the worst offenders (spreaders) out of circulation. That's all we want out of those tests. Don't confuse a "medical" test with a "public health" test. Two different things. Even a cave man can understand that, but unfortunately, the FDA has the IQ of less than a cave man.

    Your CT was 55? At that level you’ll be picking up anything , the mucosa could have had any number of residuals from other viruses even a few common cold would set it off . That’s the problem when you start testing at that level with a pcr and non symptomatic it’s a shot in the dark or if you like throwing a dart blindfolded you could hit anything .

    Considerations: 1)Some people doesn´t produce antibodies at all (2-10%) 2)Those tests are not sensitive enough 3) it may take up to 3 weeks to produce antibodies... Have you done a posterior test? Excelent explanation of lateral flow tests!

    It was a good description as to how it works. This query is sort of related to another comment below. What would the result mean if the test was too far into the future after a ‘real’ infection? If it is ‘negative’, does it just mean that one no longer has the relative antibodies? Is there a reasonable timeframe from a noticeable infection of some sort for it to produce a meaningful result? Is it true that it can’t demonstrate whether one is immune, or not to anything - except within a narrow period of time?

    sounds way better then the nasual swap things they often do. it also looks that due to it being blood, and it circulating the whole body, that the place where the infection currently might be, is irrelevant to the result.
    fo you perhaps know ehats the rate of false results?

      I'm curious about test sensitivity. I assume that the test can detect antibodies in the early post-infection phase when they are highest but I'm wondering about later time points (1-2 years post infection, for example). I know that detecting antibodies has been a challenge with SarsCov2, even in people who had confirmed infections.

    False positive. How do you know. Just because rapid came back negative? How do you prove ?

      Very good question! He did four total tests, 2 at the testing place for the PCR test, which had conflicting results, and the antibody tests both showed he was not infected. I think it's all about probability, not absolute certainty. It would be interesting to see the math behind the probability of infection based on all those test results but the accuracy of the particular tests may not be well defined, especially the antibody test which was still in development.

    Studies have shown that the sensitivity of the rapid tests are higher when using plasma/serum. Draw 100 ul blood from your finger palp and let it sit for about 2 hours. Draw 20 ul of the serum and repeat the test. Whole blood resulted in a negative test for me, repeating the test with serum showed a positive IgG result. Confirmed now with ELISA.

    One of the things I found fascinating, and perhaps it's happening in other less-studied viruses: quite a few people have gotten IgG *before* IgM for SARS-CoV-2. There was a fairly early paper from China studying factors that led to re-hospitalization, and that was one common quirk. It blew me away because I'd always heard IgG comes after IgM, but, as the old joke goes, “The immune system is very complicated ….”

    That's basically like what you have to do for a blood glucose test except you also have to add the buffer. What I am curious about is whether people who were infected might make antibodies to other surface proteins and not to the spike. Maybe a person would always make antibodies to all the surface proteins but since there are several, I do wonder if it would even be possible for someone to make antibodies to one or more than aren't the spike protein.

    Hi Vincent. Thank you for this video and explanation. Just correct the scheme because What you show in this assay as the control conjugates is not right. The control line should react with the SAME conjugates as the test line, otherwise this is not a real control.

    Thnx for the video... even though I am a scientist.. it can be hard to explain this to family members who, rightfully, feel completely lost when it comes to understanding immunoresponse, etc. It might be worth taking the time to make second video and peg your audience to elementary or junior high school level (no, seriously). I suspect most adults will find that more accessible AND it will become a resource for education. Finally, and as I am sure you are tired of doing so by now, its worth the time to reiterate that a negative test on either PCR or Ab tests is just a snapshot in time and not an indication of one's infection status after the test nor an indication of one being IMMUNE to SARS-CoV-2. I wonder how many ppl think that a positive Ab (IgM or IgG) indicates an enduring immunity (when we know that story is still poorly understood at the moment)

      It's called they have another virus. Pcr can detect a virus. It can't tell you what that virus is. It definitely can not detect disease.

    I have been getting something from Quest it is supposed a test of some sort for covid, it cost 120$

      I'm asking my PCP to order this test on Monday. Insurance pays 100%. Not FDA approved, only EUA..so should not inform any decisions re:health. Mostly just to satisfy my curiosity.

    IgM or IgG antibodies are very common in the general population and not specific to Covid, so a positive test would not prove Covid exposure/immunity.
