When sterilized media is prepared, Pour it at 40 ℃ to 45 ℃ under LAF, After solidify store plates as Inverted position. Or after test also incubate plate as inverted position. Because, 1) The inverted position does not allow the collection of condensed water droplets that could disturb the culture. 2) Inversion reduced the risk of contamination from the air. 3) The nutrients present in the media are easily distributed and available to cells in an inverted position due to gravitational force. 4) Petri dishes are easy to handle when kept in an inverted position as they are generally labeled at the bottom.
