I don't use Photoshop enough to remember shortcut keys and, unless considerate videographers like yourself include the "Image>Adjustment>Black&White" or similar in other cases, shortcut key mentions confuse the flow. I didn’t get define brush presets at all, it was greyed out. I only got define pattern. And when I did get the define brush option, the cloud wasn’t textured, it was just a solid white stamp. 😩
Check the layers panel and make sure you have selected the layer with the content you want to use for the brush, then select everything with Command or Control A. Now you should get the define brush option.
Tom You could make a smoke brush following the same technique. Just find a photo of smoke. Look for something that has good contrast and isolates the smoke against a solid background. There are lots of stock options.
Rinchen, make sure you have selected the image before you go to define brush. Check to make sure the layer is selected, and then do a Select All. That should be the solution.

Try lowering the brush opacity when you paint with it.

It’s still a solid colour with no cloud definition beyond the outside edge. :(

I hit the same problem - the brush shape must not exceed 2500 pixels in either direction
