  • Polynucleotides have been used as a medical device since 2004 but around the year 2015 they just started exploding in popularity. Here I pick apart some of the scientific studies available on the benefits of using polynucleotides in aesthetic medicine from simple fibroblast stimulation to using them as amazing wound-healing accelerators.

    I am very scared that it would add volume to my face like something like sculptra would. I know they work differently but are we sure that polynucleotides don’t add volume?

    Hi Dr. Tonks, i have seen both your videos on sculptra and polynucleotides, you mentioned that when it comes to volume, fillers are the answer. I wonder if both bio stims are diluted to promote collagen, is it possible for them to cause “overfill” look / puffy face from overly produced collagen after a year or so?

    does it help with scarring and skin tone? most of what i’ve seen says polynucleotides help with wrinkles/fine lines etc

    Thanks so much for explaining polynucleotides so well. New subscriber. Have you ever used Vitaran? I had one under-eye treatment with it 3 weeks ago. Next one due soon. I notice no difference in fine lines/crow's feet.(too early I know) I"ve tried PRP and Sunekos in the past with zero results. If anything they made things worse. When do most people notice a difference with this treatment (generally) as I don't want to waste more money on the latest treatment - it all works out really expensive, I"m not expecting miracles - a little improvement would be nice. Thanks.

    Hi Doc. Thanks for the video. I’ve got a bit of hollow under my eyes. Had filler injection in the past and had to dissolve it recently as developed puffiness where I don’t need it (a bit lower in my cheek area). Is there’s a way to correct hollowness under eyes by fillers without creating puffiness? Or only option is fat transfer,please?

      Take it out and replace it- could be in the wrong place (too superficial) or a product which holds too much water… or could just be older filler which can look more waterlogged as it breaks down. I would try again if there is a lot of hollowness. Alternatively try fibrin clot with PRP if there is a lot of hollowness. If more crepy skin then can try Sunekos, polynucleotides…

      @@DrSarahTonks Many thanks for the advice. I was thinking the same but wasn’t sure as I’m not in a beauty industry. I also think another option could be to get a few treatments of polynucleotide despite of having hollowness. Since it’s a minor hollow it could be that by thickening the skin in that area the hollowness will improve as well. Would be great to hear your opinion on this one if possible, please..

      You can get a little bit of volume from polynucleotides yes but you have to be persistent

    Hey Doc, i’ve tried the treatment and with the BAP injections technique of 5 spots per side, is there any article or discussion on how the product will spread evenly distributed on all the face? Especially does this technique ensure nucleofill to reach forehead area? I found it weird compared to papoule injections like rejuran?

    Thankyou very much..Ive spent over $10 grand just in fillers.Had profhlio done to neck .I can see some results but no where near what I was told.

      Tbh it is an expensive sport and not uncommon to spend quite a bit of money especially if you are going to someone well known. But I guess a facelift can be $70-250k depending on the surgeon

    Very interesting video, I had my first ever consultation today where I was recommended a polynucleotides procedure for my under eyes as it apparently will stimulate collagen production and could help make the skin thicker again. After seeing this video I´m not sure if this is true and will solve my issue which are dark circles and a more sunken in look, which I assume is from volume loss. I did state though that I did not want filler as I´m very scared of the over filled look especially after long term usage.

    Hello. Can i have hyaron and pn together? Can i both inject them at 90 degrees angle using 4mm needle? Or will they cause occlusion? Thank you @dr.tonks

      In theory you can yes but there would be a lot of swelling

    How interesting! Which one is best for under the eyes of a person who tends to get puffy eyes ? I know that HA has a tendency to draw water from the tissues and can puff . Does PDRN do that too ? My eyes have the beginnings of bags at 46 . This causes a tired and puffy look . It also makes my tear trough more apparent. I was told that HA fillers wouldn’t be the best idea for me due to me being prone to puffiness . I want to try having a microneedling or meso treatment with PDRN . But I’d hate to look more puffy . I have quite crepey skin under the. Eyes and the upper eyelid. Thank you ❤

      Puffy eyes and crepy skin is quite the combo. Even if you get puffy it eventually goes away when using a polynucleotide or uncrosslinked hyaluronic acid. You could combine doing this with micro needling with exosomes or RF micro needling with exosomes or CO2 laser

    Hi hun so is this correct they won’t help with sunken tear trough or lifting areas in tear trough were filler cannot be injected? As I have practioner who swears that polynucleotides will help lift please reply

      I really want to know this too! I've had Profhilo and Ellanse for my skin (both great) but my under eyes are sunken and I've heard lots of horror stories of HA puffing years later...

      I use them if someone has crepy skin and it’s not wise to go straight in with a cross linked hyaluronic acid. They are also excellent for small lines under the eye which a traditional filler won’t reach. You might get a bit of volume from them but for the best results I recommend both

    Dr. Tonks This is such an exciting product. Two questions. Can a patient get Nucleofill and ProPhilo. Would there be any benefit to that or is that just crazy. If so how would one stagger the treatments. Also, can two protocols be done at the same time? For instance the five point injections as well as the under chin or with that be flooding the cells with too much all at once? Thank you so much for all you do!

      Yes and actually this is common. I personally refer to do a whole course of Profhilo followed by a course of nucleofill so you can assess the results for both individually

      They do have both in Asia yes. Studies show parity in terms of collagen stimulation. I would say if the skin is dry and you want more of a sheen then do the hyaluronic acid. If it’s more a crepy skin issue then do the polynucleotides

      @@DrSarahTonks i did polynucleotides from Geo eyes two times until now what do you think about them ? and are there cheaper options than Teosyal because I already spend a lot of money on Platelet rich plasma and fillers and they did not gave the results I needed is it really worth it because I am struggling for a long time because I have problems with breathing while sleeping so thats why I have volume loss in first place

      I haven’t tried or heard of polynucleotides from Geo so not sure. The Teosyal redensity 2 isn’t that expensive to buy you’re paying for the person to inject it!

      I don’t know you’d have time ask him! There was a paper published in Nature which stated when comparing hyaluronic acid with polynucleotides the type 1 collagen production was greater in polynucleotides in comparison with hyaluronic acid but in any case it really is comparing apples to oranges as they are totally different and you will get something different from each. www.nature.com/articles/s41598-020-61952-w

      You’re not going to get the lovely sheen with polynucleotides that you get with hyaluronic acid and I wouldn’t say that they have as much of an effect on hyperpigmentation in my experience although I would have to check for a paper on that

      @@amardaniellok so I wanted to watch the video before I responded. I think these are all great points but it’s not really an either/or decision - clinically there is a ceiling to the improvement you’re going to get if you only do one thing- I know because I’ve seen it! So it’s better to switch between different treatments that act in different ways for the best results. I know this doesn’t apply to everyone because let’s face it, it’s expensive, but if you’re at the limit of what you can get with one modality, switch to another

      Rich PL is hyaluronic acid and poly lactic acid, Nucleofill is polynucleotides, totally different

      There just isnt anything. About 15 years ago there was a device which could be used to freeze the muscle quite literally, it had some carbon dioxide thing cooling the needles but it was only ever meant to be used on the frontalis and nowhere else. I cant remember the name of it, it didnt take off

      @@dennisguilder1Nothing lasts! You need to repeat every filler, biostimulator, treatment, whatever. I do microneedling at home with good results. Not costly at all if you do it yourself but you need to do lots of research before you start.

I'm nervous about my filler along my zygoma that it might be shifting and will make me look bloated in years to come? What are your thoughts to a more chiseled look?

    Just another waste of people’s money. Since I have stopped all these ridiculous procedures I have never had so many comments on how well I look, whereas no comments, only stares when I had Botox & fillers because they make you look ridiculous.
