it's neither heena nor Alta(paint that some are referring to in comments). in the movie this song comes during Holi celebration ( the king asks bajirao to prolong his stay and celebrate the festival with them so they could host and honour him for saving them from attack). So now the colour in her hand in this case was red gulal, gulal is primarily coloured powder with which we celebrate Holi in India. The reason they chose colour red is cause it's associated with love and affection. As in the song it's depicted as if she is requesting her lover to cover her in the colour of his love. Though the colour also help to accentuate hand movements. However literally it refers to a Gopi requesting lord Krishna to do so. I won't go into Detail of the later part as it would end up being a long comment.
