At 1:10:08 each biome in the game has one of these yellow (on the map) doors that you have to shoot to enter. They're challenge rooms that will feature increasingly difficult enemies (and often good rewards for your run), but the rooms are optional so you don't have to do them. Once you enter the room, though, you have to either win or die, you can't leave.
Enemies with red bubble shields can only be damaged easily after you shank the shield with your sword. The sword insta-pops any red bubble shield. There is an alt-fire in the game called shieldbreaker that also works, but most of the time you can get in close with the sword and immediately take them out.
Yeah, I was wondering why they weren't dying. There are also those green light things that regenerate the health of the monsters so I realize I have to shoot these first

Makes sense! I think I went 100% into these rooms but if I do not have much to lose then I would go in otherwise perhaps would reconsider.
