      @@AccreditedLaboratory SO, the main idea is that if I want to calculate LOD or LOQ for METHOD (its important) I should make fortified samples on 5 different concentrations (lowest as possible) with some replications. And then I should make an anlytical procedure with that samples. After that I will obtain results like dependency of signal from concentration (recovery should be over 70% for all samples). Then we calculating regression function and defining the linear range. The error of our regression function will be the complex error of the method. And then we calculating LOD like 3.3 times error of regression equasion devided on slope of function. LOQ - the same but 10 times. The physical sence of it that our LOD and LOQ ever would be grater than any error of method in linear (or close to that) range with recovery over 70% as it should be.

    If you are injecting 2 ppb conc. for both LOD and LOQ then how you determine which one is LOD and LOQ?

      That’s why I said to select LOQ and from that you will calculate standard deviation that will be used to calculate LOD from 3*S

    What is the guidline or referance that say that lod equal blank+3SD
    and that Sd calculated from recovery of expected LoQ

      In all guidelines you can find how to calculate LOD AND LOQ separated but you should understand how to apply in the real validation because in each you should select the lowest concentration. So, better to select the lowest conc can be quantified accurately by your instrument and method which is LOQ, then to get the S from that and apply it to LOD equation

    I think that recovery range is from 98%-102%,
    How did you get this limit (70-120%)?

      This range depend on the guideline used or as listed in the method or depend on the matrix. This range in Eurachem guidlines and in most of food residue analysis methods

    You did a mistake at the 7 minute in the video...
    LOD=mean blank+3Sd of the blank(that's the right )
    You said LOD=blank +3SD of the 6 replicate of the lowest concentration detected (that's wrong)

      Thank you for your interest. In validation you need to calculate standard deviation for all replicates then you will calculate LOD using this SD by this equation. So it should be replicated if blank sample spiked with known concentration not only blank or how you will get SD
