If you have 400 computers under the domain controllers what will you do then to enforce the Group policy? It is not feasible to log in to every single computer and enforce the GPO.

I recommend you place all 400 computers in an OU. Right click on the OU that contains the computers from the GPMC and select Group Policy Update. The enforcement should take effect on all 400 computers.
