🤗if anyone dosn't know about chromogenic substrate....so Chromogenic substrate means usually a colourless chemical, that enzymes can convert into a deeply coloured chemical. We use them, to measure if an enzyme is present. Or how much is present. We do that by adding the substrate, then allowing the enzyme to react. Then, we measure how much of the coloured reaction product, was formed.we do that by using a colourimeter or spectrometer, to measure how deep or how strong, the colour is. Sir ncert me diya hua h ki jo ampicillin containing medium me grow krenge or jo tetracycline me nhi krenge wo recombinants h or jo dono medium me grow krenge wo jo-recombinants h or hum recombinant ko select krenge kyu? Or transformants or recombinants me kya difference h? Plz sir solve my doubt

""Ab apan log waapas se ampicillin waali plate se unko nikaal lenge jo tetracycline m zinda nhi rah paae the (i.e., recombinants)".... Sir agr ham r DNA ko tet. Resistant gene pr grow kraenge to vo survive nhi kr paega or uski death ho jaegi . Aise to ham apne desired gene ko further clone hi ni kr paenge. To fir usko plating method se diff. Krne ka faida hi kya hua jb vo dead ho jaega to?? Plz sir ans fast.🙏

4:50 VECTORS help in selection of the recombinants and non-recombinants 🙄 ,but we use ELECTROPHORESIS for the selection ???Please! Do reply if u know ..🙃🌸
Okay.,, VECTORS have SELECTABLE MARKERS which help in selection of transformants and non-transformants . But ELECTROPHORESIS is used to separate the DNA fragments on the basis of their sizes. 😅😅
😅😅 they do not die 😂 the TRANSFORMANTS have the Antibiotic Resistance Genes so they grow in the medium while the non transformants do not. So they can be separated easily
RECOMBINANT DNA is the PLASMID DNA carrying the FOREIGN DNA(which we want to use ..). While the TRANSFORMANTS are the host cells after the recombinant is introduced in it. The cells which do not take up the recombinant are NON-TRANSFORMANTS. 🙃🌸🌸
Sir recombinants will die in amp antibiotic nd non rcmb survive....but we need recombinants for process as they died.so we will get no benfit...ans plz sir
Cloning site / Recognition site is the particular/specific base sequence which an enzyme recognises and act on(cut) .also called palindromic sites . Every RESTRICTION ENDONUCLEASE enzyme has its own specific pallindromic site . For exa, 5' GAATTC 3' 3' CTTAAG 5' Its a cloning/Recognition site .🙃🌸
Sir selectable markers and clonning site mein kya diff hai ? If we insert a piece of dna in tet resistance site of pbr322 then what will be the selectable marker ?
@@sangeetadevi5819 we are removing one of the antibiotic gene.... And adding another desired dna segment to rDNA.... Toh jis antibiotic resistant ko hataya... Ussi antibiotic me jb plasmid (rDNA) wala dala... Toh mar hi jaayega na... Qki uski jagha hi hamne naya sequence lagaya h
The plasmid dna in which the desired gene is inserted is known as recombinant dna. The bacterial cell that contains the recombinant dna is known as transformant.
Transformants contain the antibiotic resistance genes so they do grow in the medium containing that particular antibiotics(e.g, ampicilin ,tetracycline ..) while the non transformants do not grow .🙃🙄

But if we are introducing alien dna in plasmid at tetracycline resistant gene and it is our transformant then what is the use of selecting non transformants and transformants bcoz transformant will get killed after introducing in tetracycline antibiotic (as its resistance gene has been inactivated). So then we will only be left with the non transformants. Whats the use?!!!!😵😵😐

11:45 agr woh survive hi ni karega.... Toh transformation kaise karenge hum... Qki recombinant wala toh mar gaya na antibiotic daalne ke baad
