    Why is NaCl used in alkaline and neutralization buffers? How does NaCl play role in dna denaturation? Will neutralizing the solution not rejoin the dna strands?

      NaCl in alkaline and neutralization buffers increases ionic strength, aids in protein precipitation, and stabilizes DNA in solution.
      NaCl stabilizes the DNA double helix, making it more resistant to denaturation in high salt conditions.
      Neutralizing the solution can allow for reannealing of DNA strands, but the efficiency depends on DNA size, concentration, and buffer conditions.
      During plasmid isolation, plasmid DNA reanneals more efficiently due to its smaller size and supercoiled nature compared to fragmented genomic DNA

      maybe you should know about about the basics before commenting on it and this technique is not what is used these days.......Real-Time Quantitative PCR is used now, which you may notice is how covid is detected

      @@lumieredice485 I wouldn't say outdated, I am sure there are many specific tasks it is still used for, I just don't know. Maybe It's the same as how they teach us all the atomic theories like Thomas, Rutherford, Bohr's atomic models before moving to what is more acceptable today.
      Some prior easier, maybe outdated/faulty stuff can help in teaching the stuff that's more advanced. A bit of history lesson hidden in all the science to honor those who spent their life for it. Or the fact that we are not ready yet to understand the more advanced voodoo magic. Maybe a little bit of everything, maybe neither of it and I am just rambling nonsense. I don't know 🤷‍♀, I just like talking too much.
