The “U” side of a “cable clamp” goes over the “dead” end - the non-load end - of the cable. The “saddle” side of the clamp holds the “live” end which carries the load directly to its anchor or “origin.”
Tighten each nut back and forth until both or tight. (Check the nut tightness periodically, as materials stretch/compress slightly over time.)
And yes, have a gentle bend radius to your bent cable/loop/“bight” to equalize the load on the strands and lessen the chance of them breaking at the most acute angle apex. The teardrop-shaped pieces of steel are referred to as “thimbles.”

The wire is cable. The wire rope clamp needs to be sized and installed properly. Running cable through the conduit is fine so long as the cable is not running against the edge of the cut/end of the conduit where the sharp edge might cut the cable.
