Masculinity meant that it was how he was like a man, so you just had to talk about how he was a brave soldier turned coward, then turning into a "brave" man who fought against his enemies
I think it is significant as it foreshadows that Macduff will do to Macbeth what he did to the king - regicide- except he will be righteous in doing so. This shows how the character of Macduff basically operates as a balance to the selfish and greedy character of Macbeth, since he too is ambitious but not for personal gain, rather for his loyalty of his country and for it to have its right king (Malcolm).
