Green Kirby No, the creator of Captain Toad actually specifically said they're just friends and that Toads are asexual. Which makes sense considering some games mention Toads having spores...so just like real mushrooms they reproduce through spores, rather than reproducing sexually. So their species doesn't have sex or romantic relationships.
