This video is a parody of touhou 6, where remillia, the vampire who used donuts here, blocked out the sky with red mist, and reimu (shrine maiden) and Marisa (human magician/witch) go and tell remillia and her mansion residents to stop the mist.

This animation somewhat expects the viewer to know something about the characters and universe, so I'm not suprised you were confused. Since most of the humor comes from the characters, here's some exceedingly long context. In order of appearance, Reimu Hakurei - the first of two main protagonists, she's a human miko/shrine maiden. Her job in Gensokyo is to keep Youkai (monsters) in check and basically be a psuedo-police officer. In this game, she notices the red fog coming from the Scarlet Devil Mansion and goes to investigate. Although hotheaded and quick to start fights, she's not nearly as aggressive and borderline muderous as in this animation, it's played up for laughs. Marisa Kirisame - the second of the two main characters, she's a self-proclaimed Ordinary Magician. She is Reimu's best friend and is human like Reimu, but her magic is not divine and handed to her by a god, she put in the work to learn it herself. She's the "Funny Man" part of the duo, she's more outgoing, friendly, and chaotic than Reimu's "Straight Man" persona. Rumia - The first Youkai character. She looks small and goofy, but is actually a murderous creature of darkness who loves to devour people. Thankfully, her weak abilities and stupidity make her fairly terrible at this. Daiyousei - Originally a nameless character, her popularity made her into a psuedo-canon character. She's encountered in the same stages as Cirno, which is why she gets shoved out of the way. Cirno - Contary to Daiyousei, Cirno is a reocurring and fan favorite character. She's a hyperactive yet stupid ice Fairy. Although strong for a fairy, she's weaker than almost every other character and believes she's much more powerful than she actually is. Meiling Hong - A Chinese Youkai (though what species she is exactly isn't known). She serves as the gate guard for the mansion. She's friendly and kind, but isn't nearly as incompetent at her job as the animation suggests, which is played up for laughs. The only character she can't stop from consistently breaking in is Marisa. Patchouli Knowledge - A Youkai who lives at the Mansion due to her friendship with Remilia. She's a magician by species, as opposed to Marisa who is one by trade. She's physically frail and sickly, but her magical abilities are insanely powerful. Marisa often breaks into the Library to borrow books and not give them back, but the two of them actually are on mostly okay terms. Sakuya Izayoi - The maid of the mansion. She is assumed to be human, but there are many things about her that don't add up; she doesn't age, her eyes glow red when using her powers, she serves and is friends with monsters, and she can stop time, which in this franchise is unique to her. She's calm, proper, and sometimes severe. Despite her intimidating demeanor and scary abilities, she's fairly amicable most and doesn't often become legitimately angry. The animation portrays her as psychotic killer for laughs once again.
