Come Fly With Me - Dr. Ferguson at Fredonia
มุมมอง 1968 ปีที่แล้ว
This video chronicles the impact of Dr. Ferguson's RUI grant on the research environment at the State University of NY at Fredonia.
Microinjection into Drosophila Embryos
มุมมอง 6K9 ปีที่แล้ว
Fredonia Molecular Genetics students performing microinjection of Drosophila embryos. We fabricate the glass capillary needles and use them to inject DNA into these early embryos. The DNA gets integrated into their genome to generate a transgenic fly.

Video by Scott Ferguson, Assistant Professor of Biology at SUNY Fredonia. This video demonstrates the technique used to dissect Drosophila ovaries using tweezers.
Drosophila Media Preparation
มุมมอง 10K12 ปีที่แล้ว
Nancy Levensailor and Danielle Hindes from the Ferguson lab at SUNY Fredonia demonstrate how to make Drosophila Media. - Required Equipment - 20 quart Groen Steam Kettle (TDH-20 is shown but electric models are also available. Look on E-bay for used kettles.) - Drosofiller (59-168 Genesee Scientific) - Narrow vial trays for "Reload System" (59-207GRE Genesee Scientific) - Power drill. Any model...
Drosophila Salivary Gland Dissection
มุมมอง 111K14 ปีที่แล้ว
Video demonstrating the isolation of the salivary glands from a Drosophila larva. These glands can then be used for preparation of polytene chromosome squashes.
Can you tell me about the materials and method please?????
Bhai focus. Hi. N h sahi. S dikh hi n tha blurr h😢
Well well, there is some propaganda behind this experiment that is placed intentionally to fail students, Hail VRD
For real, never once got it and the lecturer gets mad at us for no reason like it's not my fault I don't disect larva on a daily 😢
Exactly.....There is only 10 percent out of 100 to get polytene successfully....😢hail VRD😉
Apple
I swear to god i cried so hard while doing this, my prof didn't explain anything properly and screamed at all of us for not getting polytene chromosome :( thanks your video helped me a lot, now i will try it tomorrow
murder !
1:23 👏👏👏👏👏👏💐
Good attempt bro
Delicious
Injection pressure?
Wow it's a great job
ouch
yeeeeeouch
Hey...i had a doubt...the yeast you add ....does it get completely degraded due to heat?...or else it might cause some infection
amazing!
Nice and quick
I'm in an online class because of COVID19, and I need a Drosophila salivary gland dissection in my experiment. Can I use this video? It will only be used for experimental classes.
you make this look a lot easier than it is lol nice video !
Wooooow....Thank YoU..!...
God the drosophila was still alive...
The fact that they have no PPE ..... they even wear open toed sandles ...... is an awful example to others. I would pull the video and remake with proper safety equipment.
What is the harm in anesthetizing them first? I do not appreciate your approach.
you let her know lindsay
What am I wattching
how you fabricated glass capillary
In masters we are unable to get this , nice video
Shoutout to Dr. Ferguson
wow, thanks for posting this. is the propionic acid being used as a preservative here? also, how are you controlling the PH? I would have thought there would be some variation between each batch of ingredients that would need to be corrected for?
How long does this food keep when stored?
Why don't you behead it so it won't suffer so long.
bruh....it's been dead for a while.
Oh my God.. this is much easier than holding 2 needles and pull em apart.. thank you
Yea true..😂
can we use yeast extract instead of yeast?
Yes. We use yeast extract.
I find your lack of PPE disturbing.
brutal \m/
Thanks a lot for posting this. Tried this for the first time today..killed almost 7 without satisfactory result. Now I understand it better.
Heyh
That seems so cruel to me to stretch a living animal until it breaks in half. Couldn't you have killed it first? I'm not sure if a fruit fly larva can feel pain but that did NOT look fun no matter how small he was
Many animals in nature wish for such a fast death. This was a fucking maggot!!!
It does not feel pain and it has no ability to process that pain into suffering. It's a larva, not a bunny or a cow.
fuck you
I'd love for you to see the alternate universe of what people would be doing to humans these days for therapy in medicine if we did not have the insight that we've gained via working on things like fruit fly larvae.
@@lajoswinkler I see all life as valuable, from a maggot to a human. If I can avoid their suffering in any way, I'm going to do it.
Dude you helped me alot! thank you very much!!
HI! How can i make a calibration curve log pb vs rf with this software??
What kind of tweezers are those?? I mean they're really small
look like number 5 forceps to me
This is interesting Dr. Ferguson! I will also be dissecting the ovaries of Pseudacteon phorid flies, smaller than Drosophila, in the next few days to study their fecundity, have you dissected any of such small flies?
lmao
Hi, that was very infomative. Do you have any suggestions, as to how i can shake out mature eggs from a clumped mass of ovarioles? Say by using a special media formulation and agitation by pipetting? I need to assay for number of eggs in fly ovaries and takes a long time to manually sit and tease all the eggs apart. Thanks!
Good guy. Just only 1 hit, u get the glands. Thank you. It's very helpful for me
This was extremely helpful. thank you! I did that for the first time today but my larva were much more aggressive! How did you get it to stay in a straight line once you get a hold of it from one side. mine ATTACKED the forceps holding it.
Hahahahha XD
ossmm gr8 job....ek bar me hi u took out the salivary gand...gr8 yar
Very well done. Took me many tries to become proficient at this in my lab.
We just do the dissection in PBS.
Thanks for posting this. It increased my students level of success, and I'll be introducing students to it again this coming week.
