    hello, can I ask whether it is necessary to reference a supplementary information in the manuscript? For example, if I want to publish case report of immune cell therapy in gastric cancer, the supplementary file, which is the preparation process of immune cell vaccine, is not important in understanding my manuscript. Thus I found it awkward to mention the supplementary information in the manuscript

      If you have supplementary information files posted online, the manuscript should provide a link or some notification of their existence. Why not include a statement such as "...the immune cell vaccine was prepared by Process X, as described in Supplementary Text S1."? If the process is standard, perhaps a citation would suffice, along with any modifications you made to the method.

      You should put the statement wherever it would naturally appear in a description of the processes you used in your research, such as the methods section.

      Check to see if the journal has a platform to post supplementary material along with the paper.

    I have to ask that if someone not provide the supplementary file at initial stage of communication of manuscript. Can he provide file at the revision stage?

      If reviewers need to see your supplementary file, then it should be submitted with the manuscript. There may also be an instance when an editor or reviewer suggests that a table or figure in the manuscript should be shifted to supplemental files in the revision. But follow your journal's instructions about when and how to submit such files.

      @@ScientificWritingwithKarenLMcK Thank you for your reply Ma'am. Actually, my paper is conditionally accepted but editor told me to minimize the paper size. That's why I am thinking that some of the material of manuscript can be shifted in the supplementary file. Can I move like that?

      Yes. You can move some material to supplemental files to shorten your paper. But be sure to reference those items in your manuscript.
