Is it mandatory to turn off Debugger after this testing? Will it impact end users?

Yes, absolutely turn it off before publishing 'production' content. Otherwise users will see the debug panel.

Is it mandatory to create a special playback settings for SCORM content that plays in LMS?

Not mandatory, no, but if you have tracking via Manager for some content, and tracking in the LMS for SCORM packages, it's just easier to have a separate resource so you're not having to change settings every time you switch between the two.

And the custom Continue button you used for explanation in simulation is not reflected to SCORM Cloud

It's just using a different Trainer Resource (specified in the Playback Settings). If I'd used my custom Trainer Style, my custom button would be shown here.

I have uploaded my course to cloud.scorm with debug enabled. Why would the Next button of the explanation bubble not show. It shows the placeholder for a missing graphic. Also the hotspot area of the recorded mouse action shows green, not the usual blueish color. Its almost as if a css is missing. I can't see anything in my Debug report. Any suggestions? Thanks

The Next button is contained in the Trainer Style resource. I would check that the correct Trainer Style resource is being specified in the Playback Settings you are using to generate the SCORM package. I had another client recently who had a similar problem, and the link to the help button was broken so check that and re-link it in the Playback Settings if necessary. The highlight color is specified in the same resource so that confirms this is the problem. Also make sure you choose the Include Dependent Objects when generating the SCORM package.
