I''m not sure what episode the pose was in, but the signal was being made to show where he was going actually going to toss the ball, since he was going to make it look like he was going to toss in the other direction.

The "signal" pose is him making a sign with his hand (the one that's behind his back) to tell his teammates what play they're going to be doing next. It's a standard thing in volleyball, so that the other team can't predict your moves

Once again a very entertaining and fun video to watch! I really hope that they come out with all the members of Karasuno! Even the two characters we don't see much, only one of their names was said in the anime, I'm talking about Narita Kazuhito (the other shaved head on the team) and Kinoshita Hisashi (the light brown spiky hair guy who has never played in a match in the anime so far). I grew a fondness for these two after seeing someone tell how dedicated they are to the team, they go to practice, they perform all the penalties in the training camp, they cheer on their team in the matches even though they don't get to play themselves. You see these two have the proudest face when Ennoshita does his best filling in a big role and gets to play in an official match for the first time. I hope they get their time to shine and I would love to see the whole team as nendoroids! (rambled a bit oops sorry XD)

But it's kind of hard to tell from these pictures if he's actually making the right gestures? During games he does it really subtly behind his back. It's the different signals he displaces to show the other player were, who, and when he's setting the ball.
He made these signs to represent the timing to use like a quick set to make sure the other team doesn't know what spike there going to use
