Sir if a mother transfer her share from a ancestor property she got from her father to one of her daughters through settlement and registers it ... Can the other legal heirs challenge after mother's death defending that since mother has transferred only ancestors property she got and not self acquired property??? They also have right???

Since it is a ancestor property other heirs too have equal rights. Settlement is not valid here
