Hi Aja, question about the "raw data " for FlowAI. I went through the instruction of flowAI. It mentioned that FlowAI should apply to the raw data. Does this raw data mean uncompensated data (without grid on each FCS file), ungated date (first layer of data), or uncompensated and ungated data? Thanks in advance.
The authors suggest that flowAI should be executed as the first step after data acquisition because it can help with the calculation of compensation. Definitely before gating
Hi Aja! Thanks for your amazing work. I ran FlowAI for all my samples at once, playing the plug-in in the "All Samples" group. I'm trying to export the GoodEvents files as you are showing but FlowAI keeps recalculating these new files. When I try to stop the plugging the following message appears: "This node, 'FlowAI, could not be deleted, because it contains analysis owned by a group". What can I do to solve this problem?
I would recommend exporting out the "GoodEvents" fcs files and putting them into a new workspace where FlowAI hasn't been applied to all samples. With it on all samples, I think it will calculate whenever a new sample is added (this is a guess as I've never done it this way). Hope that helps!
Thank you for clarifying different aspects of flow cytometery !
I love your videos thank you
