Microscopy: Resolution (Jeff Lichtman)

The resolution of a microscope can be defined as the smallest distance at which two small objects can still be seen as separate objects. This lecture discusses various criteria for resolution, the factors that influence resolution in the lateral and axial planes, and how to sample an image adequately using a camera or confocal microscope, such that the full optical resolution is retained.

Jeff's videos are among the best one can find here. That is why I have to comment on the example with Nyquist limit, zoom and camera pixel size. Maximal frequency of Airy function is 2*NA/lambda that is why Abbe resolution formula: resolution = 0.5*lambda/NA is more appropriate in the example. Note, that with this correction even 2x mag charger would be not enough to fulfill Nyquist sampling theorem.
