Even in the so called "Unlimited Mode" you only get to beat 7 bosses. To answer your question on how the other people get high scores, it's a combination of the Spidey's abilities you were currently running, your Team Spidey Power (aka Multiplier) and maintaining your Combo Counter. The reason the Combo Counter is so important is because anything after 39+ you will get the highest amount of points possible per every Near-Miss, Attack hit and so on. So basically you want to build your team's Spidey Power, use a Spidey with good abilities such as Bullet-Proof Spider-Man (because it's abilities are +1 to Combos from a Near-Miss and +1 to Combos from Attacks) and keep your combos going. Following the Vials during a run does provide a safe road, but it also passes up a lot of Near-Miss opportunities. The last thing to keep in mind is the Boosts & Power-Ups. There are 3 Boosts that can be used during a run, just as the name itself implies they all 3 "Boost" your score especially the "Super Score" & "Crushing Combos" Boosts and they can make a huge difference. Certain events will provide these Boosts as Progress Rewards, but you can also purchase them with ISO-8 from the Shop tab in Menu. The Power-Ups are Web Hammer, Web Magnet, X2 Stats Multiplier & Web Shield and of course you find them during a run. They do come in handy and they can be upgraded a few times with Vials, then upgraded even more using ISO-8 from once again the Shop tab. The more they are upgraded, the longer they will last during a run, but coming up with so many Vials/ISO-8 can be difficult. So there you have it, hope that it helps.
