Dulcinea and Sancho just had a bad first impression and never got along, with Sancho just generally being somewhat anti-social in general, Rodion probably kept pushing her to try and get along.

I think it’s more if La Mancha’s Don loved Bari more than Sancho as a father, which in turn let Sancho’s bloodfiend instincts return. The reason why she didn’t have any need for blood was because Don’s love was enough to allow Sancho to overcome her thirst for blood, but in here, she probably ignored her Father’s command not to drink blood, and ended up killing a weakened, thirsty Don Quixote, then took over as the Manager of La Manchaland and let the Bloodfiends there start a massacre.

But terrible all the same, because we've got an absolute horde of starving bloodfiends being set free unto P Corp.
