If you want to save space on the ESXi host, use an image profile with no tools. Please let me know if you have any questions.

I am using 7.0u3i and tried updating to 7.0u3p on a standalone host but got an error “upgrade VIB(s) loadesx is required for this transaction. Please use a depot with a complete set of esxi vibs”

In this patch USB controller will be blocked/removed from host. What if we need usb controller for VM servers servers. Is there any solution for it.

No. The patch will not remove the USB controller. It has a fix to remediate the vulnerability. Removing the controller is just a workaround until we patch to U3P
