To answer your questions in order
1. The video literally shows you that. (Though how exactly one gets molybdenum99 in the first place isn't explained very thoroughly)
2. There are measurements on the element's density and such you can look up for comparisons and notes to figure that out.
3. Saline solution, I believe.
