I think its more of a crushing defeat since Kaito(if we follow this version of the Borgia story) will be killed by random rebelling soldiers in Navarra rather than in his humiliating defeat in Italy.

To be more specific, they are expies of the infamous Borgia siblings--Kaito is Cesare Borgia; Miku is Lucrezia Borgia. (While Len bears most similarity with the figure of Alfonso de Aragon.) Here are the details: The whole story of Cantarella is loosely based on the story of the Borgias. In the video, Kaito and Miku are members of the "Vocalgia" family. They were very close as children--seeing themselves as two of a kind, but in time their feelings for each other grew into something which they knew can not be. This tormented both of them. He couldn't take it anymore, and he decided to kill Len. Despite knowing full well, that doing so would damn his ambitions and bring the wrath of a powerful family upon his head. He didn't care anymore. He hated Len, and if getting rid of him meant having to fight against all the lords of the land at once--so be it. Kaito slipped some poison in Len's drink (or used some weakening drug, so that Len could be led outside and killed by Kaito personally). Miku...was confused. And horrified. She knew exactly what was going on, but the whole scenario was so twisted and cruel--so out of hand; she was torn between the man she loved all her life, and the man she had come to love in another way. The part of her that loved Kaito was also aware that what he did had grave consequences. She feared for his life...and in time...Kaito finally paid the price for his sins. He was killed in battle, presumably with the help of Len's family, and his dark ambitions were put to an end. The Vocalgia was powerful no more. Miku survived though. Her tenacity got her through in the end and after Kaito's downfall she either lived alone or married another man out of practicality, and judging from her clothes she continues to be well-off despite the loss of her family prestige. I like this version most, especially the ending where Lucrezia/miku reaches her hand out at 1507 when Cesare/kaito died and he holds hers in 1519 when she died. The Borgias were well known for their use of poison in assasinations and the 2 siblings are suspected to have commited incest, but it's not proved to this day. Reunited in death! And such pretty art and animation.

So i was playing Assassins Creed: Brotherhood today and decided to look up Cesare Borgia (a Charakter in the game who is also a real life Historical Figure) on Wikipedia where i found out that not only is there a Vocaloid song based on him but the two seperate Mangas that are based on his life. Learning all of this has been a very surreal experience.

Though they mostly used the powder,because they mainly poisoned drinks (wines,as it was the most famous alcoholic drink in the Renaissance Italy,in which Borgias lived),and the alcohol and strong taste of the wine did a decent job in hiding the smell and bitter taste of "La Cantarella".Although,a sip was only needed,it was one of the strongest poisons.Death was inevitable and immediate.

Heres what happened.
She played, and as the soft melody of her violin enchanted the room, one stranger stood unmoved by the music. He remained in the shadows; hidden away by the mask he wore. The girl found the last note of her song. No applause came, as the crowd had not yet been broken from the spell. A pair of gloved hands silently thanked the musician. The ball continued long into the night. Though surrounded by so many dancing figures, the girl felt alone. Blue eyes stared, waiting. After her guests trickled out, she returned to her room. She undressed; sleeping only in a shift. A man in disguise finished his visit, leaving a rose for his prey. Dusk changed to dawn, and the girl woke as the first rays filtered in through her open window. This morning, just as mornings before, she discovered the bait left for her. Laying next to her was a single rose.
As she walked down the hall, she heard a song coming from one of the many rooms. The door sat ajar, as if inviting her to enter. She peeked in. The room was empty, save the piano and the man playing it. Frozen at the door, she listened as he finished his melody. Aware of her presence, he called out to her, and she entered the room. Though she had seen his face before, his beauty and warmth still shook her. He half smiled at her reaction, and she sat next to him on the piano bench. He pulled her to his lap, and placed her hands on the keys. He whispered, "Cantarella." The lesson continued in silence.
It wasn't long before the sun began to sink behind the large mountains. Dinner was being prepared, and so the two parted again. She went to the room she had been staying in, and again found a beautiful gown laid out for her. She dressed quickly in order to meet her captor. He sat elegantly relaxed in his chair. When she entered, he rose and helped her to her seat. The wine had been poured, and the girl took but a sip to his toast. Their conversation was one-sided, as usual. She hadn't spoken a word since her arrival, and yet the man didn't mind. As the night wore on, so did the drug. Her vision would blur, and she was light headed. Time held no concept to her now. Suddenly, she was surrounded by darkness. Before her body slumped over she was in the arms of the man. He kissed her forehead, and took her to his room.
He lay her gently down on the bed, despite his rising excitement. It took all his will power to restrain himself for a few minutes more. Her expression was as calm and peaceful as sincere sleep; unaffected by the anesthetic. No longer could he be restrained.
In a moment she was in a shift, and he had taken off his coat. Slowly, he traced his fingers from her face, following the curves of her body. As he leaned in closer, she let out a soft sigh from her dreams. That was all it took to stop him.
Suddenly, all their memories had flooded his mind. Laying below him now was no longer just an innocent girl. What was he about to become?
His hand met his face in shame, and a tear ran down his cheek. Was he really about to do this to her? Caught in lust and desire, the man had been blinded of the girl's true meaning to him. He realized now his faults, but could not correct them.
Through his tear-blurred vision he made out her face.
The girl woke, and saw her situation. She stared right in to the man's bright blue eyes, and spoke for the first time in his company,
“Please, capture me.”
He swiftly took her up in an embrace, and held her close to his body. She closed her eyes, and smiled. A man and a girl would become one.

so miku and kaito are siblings that grown up together and kaito suddenly realized he had romantic feelings for miku so when he saw and heard of mikus and lens engagement he got jealous and ordered the poisen (cantarella) i dont know wat he was gonna do with it but miku somewat stopped him from going yandere and spent one night with him proving she had some feelings for him too
miku and len got married and things went well until kaito (again wow) got jealous and this time he became a real yandere and killed len miku was heartbroken and saw the darkness and loneliness in her brothers eyes sooner orlater kaito had to go on a war but unfortunately he died and the ending is where miku is visiting kaitos grave.
