Please, I have a problem with the solubilization of free-base alkaloids in dichloromethane. Even if I neutralize the solution, the precipitate formed by the alkaloids never dissolves in dichloromethane. What is the issue?
Mam in the extraction we are using aq. alcohol containing Dilute acid solution. Mam is it possible that the alkaloid can form salt with the dilute acid solution?

Alkaloidal salts are soluble in aqueous alcohol Then method B aur method A me sirf itna h farak hai ke apne first step me basic solution add nahi kiya jisse ki alkaloidal salt is not converted into alkaloidal base, so alkaloidal salt remain in aqueous alcohol phase because of its solubility in it. Fir baki process same rahegi jo metbod A me kiya vahi repeat karna hai Add ammmonia so that salt get converted into free base then add organic solvent so that free alkaloidal base will suolubalized in it and 2 layers will form. One is Aqueous layer and other is organic layer Then organic layer jisme free alkaloidal base hai usko further evaporate kiya, so we get dried residue Then Dried residue ko fractionate karke(by either distilation or by doing chromatography) apne alkaloid extract kar diya am i right mam MORAL is sirf first step h different hai baki puri process same😅
