Beta-lactamases are enzymes produced by bacteria that provide multi-resistance to β-lactam antibiotics such as penicillins. These are the key player of antimicrobial resistance. Enzymes are macromolecular biological catalysts. Macromolecule means large-sized molecules and biological catalyst means these are produced by living organisms to perform and speed up a chemical reaction. Let me tell you a very simple reason to study enzymes for human benefits. Almost all the metabolic processes in the cell need enzyme catalysis in order to occur at rates fast enough to sustain life. I guess that reason is good enough to study enzymes.
β-lactam antibiotics (beta-lactam antibiotics) are a class of antibiotic consisting of all antibiotic agents that contain a beta-lactam ring in their molecular structures. This includes penicillin derivatives There are many different classes of antibiotics and beta-lactams are one of the most important example is penicillin.
These antibiotics all have a common element in their molecular structure: a four-atom ring known as a β-lactam. Through hydrolysis, the lactamase enzyme breaks the β-lactam ring open, deactivating the molecule's antibacterial properties.
Beta-lactamase enzymatic activity can be detected using nitrocefin, a chromogenic cephalosporin substrate. The bacterial supernatant is incubated with the substrate nitrocefin and by the action of the beta-lactamase the beta-lactam ring present in the substrate is dissociated and the pink colored compound is produced. The change in color indicates that the bacteria are producing the lactamase enzyme.
