24.20 it should be for public subnet you have done IGW for private subnet always IGW for public subnet for private subnet it should be nat gateway

Why did you create a database server in ec2 instance, instead of creating RDS?
Its RDS only.
For application layer and db layer subnets range were different? Didn't see it created
it's important to note that while it may not be mandatory to launch a 3 tier application in this way, it is generally considered a best practice for ensuring the security and resilience of the application. Launching the DB and application subnets in different availability zones can help to prevent a single point of failure and ensure that the application can continue to function even in the event of a disaster.

putty can be used to do from windows machine. Please make sure you create ppk from pem key..

When performing this I cannot seem to locate back to the desktop where the pem file is located, since it is just going back to the ec2 home.
Yes i have showed here how to login from ec2 to another ec2 even if your pem file is in local pc
