    Super cool video. I do have an off topic question though. What do you do before you sit down in front of a flow hood? Bathe? Clean clothes?

      Nothing. The whole idea of the laminar flow hood is to blow stuff away from the work area. SAB is a different story. You should clean and alcohol everything before it goes in the box. But with a laminar flow hood, the technique is what is important. Never work 'in front' or over plates, etc. If you watch my videos closely, you will see how to do it. I hate to say it, but it is something that comes with experience. I cringe sometimes when I see what others do in their videos. Wear tight-fitting gloves, lots alcohol on all surfaces, keep clean stuff upwind of you, etc. Biggest thing! Don't get discouraged. Contamination happens all the time, but if you consistently get it, something is wrong.

    Hey Ed, I have heard you say something before about a mono being able to be "sudo rhizo" . If possible could you display a mono that shows those characteristics? I'm pretty sure I have some cultures that might be doing that.

      I don't have any images handy. Pseudo-rhizomorphic is just rhizomorphic-looking growth that doesn't have clamp connections. Rhizomorphic growth almost always indicates a dikaryon in our babies, but very rarely, it can be monokaryotic. You need a scope to be certain.

      @edwardgrand Thanks for the reply! I have a Amscope I have been messing with. Was just a tad confused when I thought I had a mono verified by lack of clamps start growing rhizomorphic. I even took another slide of it after it changed, and it still seems to lack clamp connections. Guess I'll try to match it with some others and see what we get! Thanks again Ed!!

      @Snufulufugus it's hard to know without a photo. It really depends on media too. But if you don't see thick, heavy rhizo, and no clamps, should be mono.

    I thought an axiom of science was that you cannot prove a negative. I'm not saying you can't be fairly certain you don't have a clamp connection.

      Cubenis dikaryons are rhizo on the right media at the right concentrations.

    8:50 This is why I'm focusing on building a grain spawn business. Everyones focused on selling genetics right now. In the meantime, I'm working with all these genetic slingers who can't be bothered to process lots of grain at a time :) :)

      Other than sterility, proper grain prep is the most important thing in this hobby. It took me years of experience to figure this out and I still F it up frequently. So many variables in a new batch of grain. Now I know what all those 'food scientists' get paid for! TBH, most people can't cook pasta these days, so you might have a large market. Good luck.

      @@edwardgrand I have materials testing experience so thats a huge plus. One of the easiest tests to run is a gravimetric moisture analysis and, as you know, getting the moisture levels right is definitely a huge factor. Having good suppliers and buying large batches is pretty key too, I've found. Thanks man! Take care out there. Lovin' your videos.
