If you're used to organic media, then one or two layers is actually extremely easy to do. Especially in programs that, to a point, mimic organic media.(Such as Paint Tool Sai and Corel Painter)

I've change my marker setting like you through your setting you posted on DA but I hardly control it because the colors that I draw seem much more lighter than the one I choose?? Plz help :(
When you open SAI I usually go for where it says 'presets' and choose 'A4 - 350dpi (JIS) and it creates a rather large canvas. I just zoom in 50%, draw what i need to in that space, and crop to the drawing.
@Kaqome I know you're asking Ru3na, but from my experience it doesn't really matter too much on a normal painting, however once during my final exam for Art i had it up at 96% cos of the amount of detail in my work and the number of layers I had (i regret to say it was over 200) and it rendered me unable to save the image in SAI form, I had to save it in PSD. So unless it goes up to the 95+% range and you have over a hundred layers I think you should be safe(:

the marker tool is a very versatile tool in paint tool sai and personally its one of my favorites. And this artist here knows her stuff about it.

Hi how do you get bold colours because everytime i draw my colours always look transparent although the opacity is full
