When it makes that sound and it glows you're supposed to press "RT and LT" or "RB and LB" to do a special. If they're blocking it won't work and you can only do it once.
Edit: I'm not really good at the game I just played it as a kid.
If you do go back to this when you have that red aura you'll be able to do your rage art which shows how to do it during the loading screen and its basically their special move

That L1 is a instant combo button. Hold it and press one of the buttons and it will do that combo
The glowing means you can use your rage arts which deals a lot of damage as well as your super moves

And you can just press the right bumper to use your rage drive it's like a finishing move

@@stragalot0 Only for some characters. There are a lot of characters who are easy to use. Like Eddy Gordo is one of the easiest characters to use.
