Hi there! 👋🏻 We live on a 1-acre farm right in the middle of the busy city of Phoenix, Arizona! Our names are Kevin & DaNelle, and we moved here in 2009, when our kids Ethan & Lydia were just 6 & 4 years old 🥰 Over the years we've raised goats for milk, chickens for eggs, built a fish/swim pond, planted fruit & nut trees, and grew a large garden full of veggies. We've also raised a bunch of other animals like pigs, sheep, turkeys, geese, peacocks, and more!
Now that our kids are away at college, we have settled into doing what we love most -- which is raising & breeding goats ethically to improve genetics. But don't worry, we'll forever love our goofy pet goats like Luna the herd queen & Willow the stump lover. We also still love raising chickens for eggs, gardening, tending to the fruit & nut trees, and growing fish in the pond. We use the fresh goat's milk to make cheese, butter, yogurt -- and we use that + everything else we harvest on our little farm to make delicious meals. Having a getaway from the busy world right in our backyard brings so much joy to us each day, and we hope it brings some to you, too. 🥰

It's fun watching this as an embryologist. The struggle with getting straws out of the tank is too real lol (sometimes i just grab it with my bare hands, we don't wear gloves bc there used to be a chemical in the gloves that embryos were sensitive to)
Also, the issue with the dead frozen sperm could be a few things. It might be that that buck's sperm just don't freeze well, as it can vary drastically between individuals. It also could be that the straws thawed a little in the process of transport (I don't remember how you got the straws so this might not be true). But thawing and refreezing even a little can be devestating for sperm. This is just from my experience in bovine but I've never worked with goats so it might be different :)
@@sokibuscus4998I learned from my experience in the equine industry that different extenders can have an effect on the freezing success for different stallions, so I would think that probably applies to bucks (and maybe bulls too).

Hello! You did a good job! My dad was a very early AI expert and I spent every Saturday from 1968 to 1978 with him watching him AI the best Holstein cows in the world. He worked with Carnation Genetics and he was a dairyman and consultant on proper cow nutrition too. I'm so anxious to see the babies River has. Hopefully she has at least three! I think you should pull another buck straw and check it, then rebred her.

I would be very curious if the facility that collected Moon Station checked the motility and mobility of his sample before sending it out. We had to do that for any “samples” we collected when I worked at a horse breeding facility. Before freezing, cooling, or fresh using, we checked every single sample. If it was less than 50% mot/mob then we made a note and checked the temperature from the previous 20 days.
They do check it first, it was 67% motility so it may have been when I transferred it:)
Oh, and: I'm really glad you posted such an in-depth video, this really doesn't look THAT hard, outside of all of the equipment needed for it. It looks kinda fun, to somebody who likes to tinker and collect fun science stuff! Gotta say, it was a mistake to watch the...heck. What's the camera called, I've forgotten. The drain snake camera 🤣while eating a bowl of soup, but listen, that's my cross to bear.

Back in the early 80s, my friend and a mentor started using AI with a herd of dairy goats. It was challenging but they were successful. Togenbergs and Alpines.

I’m surprised. Thought you talked daybeds or Murphy beds for the main area! No matter what I’m sure it will look great though!
Sperm can die within a few minutes in the air. It truly depends on temperature/how you prepared the slide and stuff like that. In the air it simply dries out and dies because of it.
I think if you were to add 9 gram kitchen salt (NaCL) into about a liter of water, you might be able to add that to the slide as well as the drop (probably a drop of it) you might thin out the amount of sperm you see but keep the sperm a bit more wet and it should perhaps survive a bit longer. (Note - I work in a Laboratory, this is what I remembered of Biology, work in Chemistry)
That being said bad straws excist! Just a fun fact for if you have less time and need the slide to remain ok. Another option might be putting the slide in the refrigerator (as keeping samples cold for some cases can help) although I never tried thát before. It might work it might not work...the last part I mentioned about keeping cold would have to be figured out by trial and error😅
I didn't use cover slides on samples because they hadn't arrived in time, so I'll def use them in the future. But regardless, there was still a big difference between the two bucks' samples. So I think the problem was with the semen and not an issue of them drying out on the slide. But yes, all these details you have to be sure to think of -- it's surprising how fragile it is! :)

You would be surprised. In laboratories there is one temperature, between 68 and 77 F. Just to ensure that the temperature does not give us faulty results. Not to talk about humidity that also influences it. So two of the same tests, on the same day, with different temperatures/humidity can cause different results!
That being said having multiple straws turning up badly will suggest something is going on.
Laboratory work is quite difficult and like I said if you need tips or tricks. Just let me know ;)
Did you know that instead of covers you can also just use another slide and put it on top? Oh and if you really zoom in a lot and the lens might touch the slide, use some oil! It protects the lens! ;) You can clean the lens afterwards by using a bit of a soft kind of wet wipe.
To prevent air from being added between the cover slide and slide, you can actually use tweezers and very gently from left to right put it down. It should keep the air in the sample to a lower level! Like you pick it up with tweezers, lower the left side down and than carefully press the right side down as well. Otherwise you have to pick it up in a corner with you fingers (so around it not touching the glass itself, just the corner of the glass) and drop it real fast in place.
The cover slides break very easily! So if you have trouble with them breaking, just use a second slide and drop it on the first in the same way. We done that at Microscopy often enough if the tiny, sticky cover slides were giving too much trouble.

If you have just one drop of sperm instead of trying it with the cover slide immediately perhaps try a drop of water out of your swimming area in your backyard. It might have some fun little organism to look at and it can be practice to use slides. If you have no second drop and you have to redo the slide making it is a lot harder to have good results!

Never drop the oil on the lens by the way, always on the slide..
And if you microscope has two parts that you can turn to make it zoom. The smaller one is slower, the bigger one is faster. Never use the bigger one for the last bit of zooming. It can hit the glass under it and actually damage the lens!
