There are a lot of programs that have built-in or downloadable plugins that are meant to isolate vocals. For example, a free program 'Audacity' has a built-in plugin called something like 'Isolate Vocals' and it just detects things that aren't voice, and remove it. I assume (s)he just downloads a video from youtube or audio file of the song, opens it in something like Audacity, and then uses those plugins.
