That's called a pinch harmonic. Look up here on TH-cam "how to do pinch harmonics" or "pinch harmonic tutorial". There are several good videos explaining how to do them. They are a bit tricky to do in the beginning, but keep practicing!

I think for this video I just cut some of the mids (song is already very scooped for mids), and added a tad more mid-range on my own guitar to make it fit nicely in the mix :)
Yes that's what i'm not sure of. I don't know that. Just tune to regular standard E tuning, tune everything a whole step down then tune the thickest string another step down.
