If memory serves you had to rush Spaceways on hard difficulty, which was all about killing nothing so enemies couldn't spawn at some point. That might be the very last character I unlocked, I even thought I had unlocked all of them before I got her.

Sad thing is, out of all the bizarre characters (Duckman Drake, Gingerbread man, Robofish) she was the only one not to appear in more than one game.
