    Great video Mr. Mayne. Very good presentation. A few suggestions. 1. For gelling agent you can use - KIRKLAND SIGNATURE NATURAL PSYLLIUM FIBER 2. If you add 5 mg/L of Copper Sulfate Pentahydrate (buy from Home depot or such stores) - (You take a 5 grams dissolve in 1 litre to form Mother solution and add 1 ml of it to your 1L media). you will have more shoot and root development and less chances of fungal, bacterial infections.

      @@AquaMayne Not only that, since CuSO4 kills off any Fungus or Bacteria it diverts more energy to shoot and root multiplication. In most plants (value of best ppm differs) the number of shoots and roots is almost double when compared to no copper used control bins. You can look up some examples of Banana tissue culture and others in Google search.

      @@bijanmozafari3826 Most of them gets benefit from CuSO4.5H2O. It is better to use 1 ppm to be on safe side. Do not discard remaining solution in small water bodies as it can cause microbial life to collapse.

    Enjoyed you video. What is the protocol to get the multiplied and rooted plants back into the aquarium? First a day is sterile distilled water?

    Great video man! it's a fun experiment, but I think it is too much work for just a common plants..
    what you should tissue culture are many bucephalandra species, aponogeton madagascar, and anubias.
    Those are much more rewarding for a tissue culture propagation

    hello, if i want to propagate a plant like java fern or anubias, do i use a piece of rhizome or a piece of leaf and put it in the media

      Rhizome would be more guaranteed but you can do a leaf if you use a medium with rooting hormone.

      No If you do it in a still air box or laminar flow hood you can transfer straight over very carefully

    Is there any special procedure for aquatic plants? Do they need to be transitioned out of water in stage 0 and do they need to be transitioned into water in the final stage? Do they do just as well as regular plants in the multiplication media?

    So you make one set of media cups with multiplication media, and then transfer the plantlets into cups with the rooting media?

      Yes that is the basic order of TC. Some people skip rooting though. Obviously you will not have as many roots for the plant to establish outside of TC though if rooting is skipped.

      Ross Tafarian yes I made AR red calluses. I failed at multiplication after callus though

      Most lived and added to the aquarium, my most recent vid is a laminar flow hood for TC in the future when I get time :)

    Merci pour cette vidéo qui va bien m’aider même si du coup votre méthode n’est pas entièrement stérile je vais utiliser le même procédé mais avec une glowe box faite maison afin d’être sûr de travailler en milieu stérile
