In my previous semester I had a control systems course, and laplace transforms was the only way to solve any system in a reasonable amount of time

From my guess, he seems to be holding the pen like someone right handed would be, but the camera shows him using his "left" hand so most likely it's a camera trick

You ABSOLUTELY can use laplace transform WITHOUT initial conditions you'll just get the general solution for your differential equation.
To do that you'll just have to factor the y'(0), y"(0), ... as constants.
