hello, I saw this build and i wanted to try it but im not sure about levelling. I just go lightning arrow ligtning rod till I get poisonous concoction ?

I levelled with lightning arrow/rod personally but I'm sure other routes work as well (poison, etc). Good luck and have fun! :)

how do you keep the mana flask charges up when fighting bosses?

Anything with Flasks Gain x Charges per Second or Mana Flasks Gain x Charges per Second combined with Increased Flask Charges Gained gives pretty good passive recharge for Boss fights in case they go long. I have 0.17 on belt, 0.27 from Alchemist's Boon (30 Spirit), and 0.1 for mana flasks from Arcane Alchemy in the Passive Tree. Arcane Mixtures in the Passive Tree also offers 0.1 for mana flasks, but I do not go out to that node
