Can we apply diversity index for just 2 species of 3 different locations? like can we find evenness and richness of 2 species from 3 different locations?
If there are two species in a place, then the richness is two. Evenness requires counting the number of individuals in each species and comparing them to one another. Most biodiversity indexes require both a count of the number of species and a count of the number of individuals. If you have this information you can determine the biodiversity of as many different locations as you desire.
The visual appearance of this video makes it look dated (as of 2020 when I post this comment.) The actual material of narration is just a relevant as ever, though.
