When I watched that episode of Supernatural I never thought Misha was an asshole, I just thought he was very funny and a great sport to be playing someone like that who was supposed to be him. It was that episode that made me start watching TH-cam footage of the actors rather than just fan-made vids of the show, and I've laughed so hard at all these convention videos! Jensen, Jared and Misha are so so funny.

In the episode French Mistake which is in season six Dean and Sam get sent to an alternate reality where they are stars in a TV show Supernatural which is where Misha gets to play Misha as he isn't Castiel in that alternate reality, but instead plays Castiel. That's what they're talking about.
