I would suggest doing some research to see what type of master degrees are out there that have programs that focus on youth theatre/educational theatre. Just make sure the training you are getting is focused on the job you want to eventually get.
Typically, yes they do. Even adjuncts need to have minimum a master degree now and some experience in their field professionally. Adjuncts though will often be a gateway to a full time position as many colleges often hire from within. If you have no teaching experience at the college level then working part time is a great way to build your resume and learn how to teach along with crafting your own pedagogy. Note: There are (sometimes) exceptions depending upon the school, position and other factors relating to the class/s they are needing to hire someone to teach.
